Is it possible to set up a series of shortcuts on one machine and then use it on another machine? I want to do this for my work computer which is locked down so cannot install the drivers on it.

No, that will not be possible. You will need to have the Xencelabs driver installed on the computer that you will be using the Quick Keys or Pen Tablet with.
